Comparison of Iiposome-incorporated antibiotics and free drugs in inhibition of L. pneumophila multiplication within U937, human monocyte-like cell line


Abstract
The facultative intracellular pathogen, legionella pneumophila, multiplies within and kills human blood monocytes and alveolar macrophages. The therapeatic efficacies of liposome-incorporated antibiotics against L. pneumophila were evaluated in a model of intracellular infection. ¥â-lactams are very effective(ampicillin MIC=0.63§¶/§¢) in vitro against L. pneumophila, but clinically are not useful. It¢¥s mainly because of their poor penetration properties into cells. Liposome-incorporation is an useful delivery method for antibiotics to macrophages and monocytes. In this experiment phospholipid was composed of DMPC : CHOL : DCP=7 : 1 : 2 molar ratio. Compared with free form (MIEC=36§¶/§¢) against intracellular L. pneumophila. And Ciprofloxacin also showed improvement (about 8 times less MIEC).
